Аннотация:ObjectivesGli-simior1 is a Gli-related Kruppel-like zinc finger protein that is composed of 660 amino acids containing a bipartite nuclear localization signal and a zinc finger domain consisting of five tandem zinc finger motifs.Glis1 participates in the development of the craniofacial region and promotes direct reprogramming of somatic cells during the generation of induced pluripotent stem cells generation.Since it is considered that the process of carcinogenesis is regarded as the process of reprograming of somatic cells to cancer stem cells, we hypothesize that Glis1 is concerned with carcinogenesis of oral mucosa , and analyze the expression of Glis1 in epithelial dysplasia (ED) and squamous cell carcinoma (SCC) . MethodsFour cases of normal mucosa, 44 cases of ED and 249 cases of SCC were examined by immunohistochemical analysis.The quantity of mRNA of Glis1 in normal epithelium (NE) , ED and SCC were evaluated using laser microdissection and real-time RT-PCR. ResultsGlis1 was localized in the cytoplasm, but not detected in nuclei in the suprabasal to cornifi ed layer of NE and ED.In contrast, Glis1 was expressed in nuclei of neoplastic cells in ED and SCC.Glis1 mRNA expression was increased in ED compared to NE and SCC. DiscussionNuclear translocation of Glis1 plays an important role for the process of carcinogenesis of oral mucosa.It is suggested that the disturbance of control of intracellular localization and abnormal degradation of Glis1 may occur in oral dysplasia-carcinoma sequence.
